For some reason the 'increase' car setup preparations button isn't working for me. I can see it's only available on a Monday and Tuesday, but clicking the button does nothing, except refresh the page - no error message appears.

I tried it today at 6;30am UK time and yesterday at 11:30pm UK time (currently GMT+1 during summer) with the same outcome.

The button did not work for me during week 3 either, but during week 4 it DID work?

Is there also a certain timeframe window during Mondays & Tuesdays, that the simulator is 'open'? Or is this a glitch/bug?

Maybe. It was very late on when I tried last night so may have crossed over into Tuesday game-time.
